    Recording single-channel activity of inositol trisphosphate receptors in intact cells with a microscope, not a patch clamp.

    Optical single-channel recording is a novel tool for the study of individual Ca2+-permeable channels within intact cells under minimally perturbed physiological conditions. As applied to the functioning and spatial organization of IP3Rs, this approach complements our existing knowledge, which derives largely from reduced systems - such as reconstitution into lipid bilayers and patch clamping of IP3Rs on the membrane of excised nuclei - where the spatial arrangement and interactions among IP3Rs via CICR are disrupted. The ability to image the activity of single IP3R channels with millisecond resolution together with localization of their positions with a precision of a few tens of nanometers both raises several intriguing questions and holds promise of answers. In particular, what mechanism underlies the anchoring of puffs and blips to static locations; why do these Ca2+ release events appear to involve only a very small fraction of the IP3Rs within a cell; and how can we reconcile the relative immotility of functional IP3Rs with numerous studies reporting free diffusion of IP3R protein in the ER membrane

    Surgical site infection (SSI) occurs when a wound created as part of a surgical procedure becomes infected. SSI is one of the most common healthcare-associated infections and occurs in approximately 5% of patients undergoing a surgical procedure. SSI may lead to patients suffering considerable morbidity or mortality and have significant cost implications. The aetiology involves the interplay of host, environmental and pathogen factors all of which should be addressed in seeking to reduce the risk of developing an infection. The presence of prosthetic material reduces the number of bacteria necessary for an infection to develop and can give rise to treatment and diagnostic difficulties. The responsible organisms are most commonly Staphylococcus aureus and S. epidermidis. Diagnosis is frequently problematic and antibiotic treatment alone is often ineffective due to biofilm formation necessitating removal of prosthesis in many cases. Prevention of infection is by far the most important aspect of prosthetic implant surgery. Patient optimization is equally important as the cutting edge research into biological prostheses in reducing the incidence of prosthetic infection in future practice

    An analysis of HCN observations of the Circumnuclear Disk at the galactic centre

    The Circumnuclear Disk (CND) is a torus of dust and molecular gas rotating about the galactic centre and extending from approximately 1.6pc to 7pc from the central massive black hole, SgrA*. Large Velocity Gradient modelling of the intensities of the HCN 1-0, 3-2 and 4-3 transitions is used to infer hydrogen density and HCN optical depth. From HCN observations we find the molecular hydrogen density ranges from 0.1 to 2 ×\times 106^{6} cm−3^{-3}, about an order of magnitude less than inferred previously. The 1-0 line is weakly inverted with line-centre optical depth approx −-0.1, in stark contrast to earlier estimates of 4. The estimated mass of the ring is approximately 3 −- 4 ×\times 105^{5}M⊙_{\odot} consistent with estimates based on thermal dust emission. The tidal shear in the disk implies that star formation is not expected to occur without some significant triggering event.Comment: 17 pages, 8 figures, 7 tables (including 3 landscape tables generated with 2 separate Latex files

    Developing preparedness for flexible delivery of training in enterprises

    On a basis of research and literature review, Smith, in 2001, suggested a model for the development of preparedness of learners and their workplaces to support the flexible delivery of training in enterprises. Using the model as a framework, he then developed a detailed set of strategies that may be used in operating workplaces to develop learners and workplaces for effective flexible delivery. The research reported here was designed to test that strategy set in 12 different enterprises to assess the feasibility of their implementation in operating workplaces. The research shows that a majority of suggested strategies are feasible for implementation; some are feasible with qualification; and a minority were not seen as feasible.<br /

    Determining the quality of mathematical software using reference data sets

    This paper describes a methodology for evaluating the numerical accuracy of software that performs mathematical calculations. The authors explain how this methodology extends the concept of metrological traceability, which is fundamental to measurement, to include software quality. Overviews of two European Union-funded projects are also presented. The first project developed an infrastructure to allow software to be verified by testing, via the internet, using reference data sets. The primary focus of the project was software used within systems that make physical measurements. The second project, currently underway, explores using this infrastructure to verify mathematical software used within general scientific and engineering disciplines. Publications on using reference data sets for the verification of mathematical software are usually intended for a readership specialising in measurement science or mathematics. This paper is aimed at a more general readership, in particular software quality specialists and computer scientists. Further engagement with experts in these disciplines will be helpful to the continued development of this application of software quality

    Sex differences in Cognitive Abilities Test scores: a UK national picture

    Background and aims. There is uncertainty about the extent or even existence of sex differences in the mean and variability of reasoning test scores ( Jensen, 1998; Lynn, 1994, ; Mackintosh, 1996). This paper analyses the Cognitive Abilities Test (CAT) scores of a large and representative sample of UK pupils to determine the extent of any sex differences. Sample. A nationally representative UK sample of over 320,000 school pupils aged 11-12 years was assessed on the CAT (third edition) between September 2001 and August 2003. The CAT includes separate nationally standardized tests for verbal, quantitative, and non-verbal reasoning. The size and recency of the sample is unprecedented in research on this issue. Methods. The sheer size of the sample ensures that any sex difference will achieve statistical significance. Therefore, effect sizes (d) and variance ratios (VR) are employed to evaluate the magnitude of sex differences in mean scores and in score variability, respectively. Results. The mean verbal reasoning score for girls was 2.2 standard score points higher than the mean for boys, but only 0.3 standard points in favour of girls for non-verbal reasoning (NVR), and 0.7 points in favour of boys for quantitative reasoning (QR). However, for all three tests there were substantial sex differences in the standard deviation of scores, with greater variance among boys. Boys were over represented relative to girls at both the top and the bottom extremes for all tests, with the exception of the top 10% in verbal reasoning. Conclusions. Given the small differences in means, explanations for sex differences in wider domains such examination attainment at age 16 need to look beyond conceptions of `ability'. Boys tend to be both the lowest and the highest performers in terms of their reasoning abilities, which warns against the danger of stereotyping boys as low achievers

    Beyond supply and demand: addressing the complexities of workforce exclusion in Australia

    Workforce exclusion is a complex and enduring problem in Australia, with some groups of job seekers more likely to be disadvantaged in the labour market than others. We identify a dominant unemployment narrative of ‘work first’ that surrounds unemployment interventions, and ignores the nature of disadvantage and its relationship to workforce exclusion, and reduces unemployment to a simple matter of labour market supply and demand. This approach privileges immediate economic productivity and exit from welfare payments over sustainable attachment to quality jobs. We examine fourteen programs for disadvantaged job seekers under one national provider network. Data was gathered from eleven semi-structured telephone interviews and eight evaluation reports and analysed using thematic analysis supported by NVivo. Our findings challenge the dominant narrative and argue that both ends of the supply and demand equation need to be examined, stressing the importance of a partnership-orientated and capacity building focus on the unemployed person, and the significance of quality employment with long term support. We identify the importance of acknowledging job seekers’ strengths, aspirations and preferences, and of job seekers having agency to determine their own pathways with support from service providers
